Abstract

Anti-Mullerian hormone (AMH) secreted by Sertoli cells in the male fetus leads to the regression of the female-specific Mullerian
duct. In this study, our aim was to determine and statistically examine the link between the activity of AMH and the apoptosis of the
Mullerian duct. We made use of 13th and 15th days old male embryos (E13 and E15) obtained from Balb/c mice in the study. TUNEL and
Hoechst 33342 labeling was performed in order to distinguish apoptotic cells within the Mullerian duct. For the statistical analysis of
apoptosis, we used the Stereo Investigator program and calculated the number apoptotic cells within 1 mm3 of the Mullerian duct. Student t
test (95% confidence interval) was applied on the data we acquired. In E13, when the AMH just starts being produced and the Mullerian
duct completes its differentiation, we did not come across to any apoptotic cells on the many section layers crossing the Mullerian duct. In
E15 where AMH hormone synthesis has finished and the Mullerian duct is being broken down intensely, we have found a significant
increase in apoptotic cell numbers. Apoptotic cell density per 1 mm3 of the Mullerian duct was determined as 0.358 in E13 and as 1.438 in
E15. The difference was determined to be statistically significant (p<0.005) within the confidence interval of 95%. This study points out
the importance of AMH-mediated programmed cell death and hormonal alterations; two major events taking place during the regression of
the Mullerian duct and the determination of gender in embryogenesis.
