Alabalıklarda Sekans ve PZR-RFLP Analizi ile Tür ve Orijin Belirlenmesi Üzerine Çalışma

Özet

Genetic studies have shown a rapid increase in recent years. In parallel with this increase, genetic tools can be applicated in many different subjects in aquaculture. In this study, some methods has been given that are applicated in aquaculture. Beside, in this study on trout species, two types of genetic methods (sequences and mtDNA-RFLP) were used for different purposes.On Exports and imports of fishery products, it has been given importance monitoring and control, which species / origin, and even made compulsory. In this context, in our country imports of some fishery products such applications are encountered. Similarly, it was applied for to be imported fish fillets for determination of which species. DNA was extracted from frozen fillets according to phenol / chloroform method. 16S and cyt-b genes were amplified with PCR. According to sequence analysis results of fillets 99% similarities was determined with the S.salar species.Another study was carried out to determination of species and origin of stock of a farmer that want to breed. Sampels were collected from fish farm. DNA was extracted from fresh muscle tissue according phenol / chloroform method. ND1 and cyt-b/d-loop genes were amplified with PCR and digesion with different () restriction enzymes. Determineted haplotypes were compareted other studies which previously concluded that “Determination genetic of of Turkey brown trout populations genetic Structure Project” is evaluated. As a result of these comparisons genetic origin of fish was found to Abant region and as subspecies is S.trutta abanticus. Nature trout is as a genetic resource is important species. So we recommended that tissue and DNA bank, and genetic data (including different methods data) and available database should be established. As a result of these this study will further detail.
