The bacteria used in this study, with lipolytic activity, were isolated from the soils, collected from around the olive orchards and olive oil factory, and from
raw and pasteurized milk, using Tributyrin Agar media. The Gram reactions of the isolates were determined and the gram positive isolates were selected and
identifi ed to determine the lipase activity.In addition the Tributyrin Agar media, the lipolytic activity of the strains were determined under visible and U.V. light using Rhodamine-B Agar media.The quantitative lipase activity of all gram positive bacteria were determined with the spectrophotometric method using p- nitrophenyl- palmitate as a substrate. In 9 isolates from soils did not show lipase activity. The lipase activity of the other isolates were determined between 2 U/ml and 581.5 U/ml. The maximum lipase activity showed in Staphylococcus sp. NS 02-1. In 7 isolates from milk did not show lipase activity. The lipase activity of the other isolates were determined between 1.6 U/ml and 564.6 U/ml. The maximum lipase activity showed in Staphylococcus sp. RB 07-2.
