Tümör Nekroz Faktör α (TNF- α)’nın Hipoksik ve Normoksik Koşullarda Hepatoma Kanser Hücrelerinde (Hep3B) Sitotoksik Etkisinin Belirlenmesi

In the present study, the antitumor effects of tumor necrosis factor alpha (TNF-α) was investigated in human hepatoma
cells, Hep3B under normal oxygen and hypoxic conditions. Cytotoxic effect of two different concentrations, namely 50 U/ml
and 500 U/ml, of TNF-α was determined at three time points, 24h, 48h and 72h under normal oxygen and chemically induced
hypoxic conditions and followed by MTT test. The results were statistically analyzed by one way ANOVA test. The dose and
time-dependent antitumor activity of TNF-α was determined in Hep3B cells. In normoxic conditions, 50 U/ml and 500 U/ml of
TNF-α on Hep3B cells exhibited the proliferative effect although no effect was detected at 48 h and 72 h compared to control
group. In hypoxic condition, similar to proliferative effect in normoxic conditions, 500 U/ml TNF-α caused the same
proliferative effects at 24 h and 48 h. In hypoxic condition, 500 U/ml of TNF-α led to statistically significant cytotoxic effect
of Hep3B cells at 72h.
