Özet

In this study, previously phenotypic reactions’ identifed 7 tomato lines against fungal pathogen, Alternaria solani Souer, were inoculated with
bacterial speck disease pathogen Pseudomonas syringae pv. tomato (Pst) DC3000 isolate to investigate resistance reactions. Bacterial Pst DC3000
isolates were grown in King B broth and the bacterial concentration were prepared and then were injected with intercellular area of tomato leaves.
Bacterial growth was monitored during 3 weeks, and plant symptoms were assessed with pathogenicity tests. Therefore, from inoculated plant
leaves 1 cm diameter discs were cut and macerated with MgCl2buffer at 0., 7., and 15 days post inoculation. The bacterial containing liquid was
diluted and 20 ml sample was dropped with three replicates on to King B agar. Bacterial colonies were counted at 24 hour after incubation and
bacterial concentrations were identified in each leaf to figure bacterial growth in the leaves of 7 tomato lines. The pathogenicity results revealed
that EBR1 and EBR5 tomato lines were most susceptible, EBR2 and EBR3 were susceptible, EBR6 and NC84173 were resistant and EBR4 line
was the most resistant tomatoes.
