Sentorya (Centaurea tchihatchefii Fish.&Mey.) Endemik Bitkisinin Çoğaltımı Üzerinde Çalışmalar

Centaurea tchihatcheffii Fish.&Mey is critically endangered endemic species which grows on limited area around Gölbaşı-Ankara Province in Turkey. Propagation of this species has remained low in its natural habitat due to high seed dormancy due to embryo and needs a long vernalization to break it. In this research, the propagation of this species investigated in vivo and in vitro conditions. Besides, to obtain poliploid plants, colchicine treatment has been done in vitro conditions.
In the germination studies, to break of the dormancy, GA3 and stratification applied to seeds. The highest germination rates (58.89 %) obtained from the seeds applied GA3 and stratification for 4-5 months. In vitro germination studies, the highest germination were maintained on MS media supplemented 1 mg/L GA3 or 2 mg/L GA3 and the germination rates were determined as 83.6% and 85.2% respectively. The highest shoot formation was maintained on medium supplemented with 0.5 mg/L IBA + 0.5 mg/L BAP at the first subculture and the number of shoot per explant was 2.05. The best rooting was achieved on hormone-free MS medium and the MS medium supplemented with 1.0mg/L IBA. Their rooting rates were 70 % and 86 % respectively. C. tchihatcheffii has been propagated through seeds naturally and through in vitro techniques. Consequently C. tchihatcheffii has a high potential as outdoor ornamental plant.
